Description
TCI D5702 DMT-dT Phosphoramidite is a thymidine phosphoramidite monomer used for the chemical synthesis of DNA oligonucleotides. Its 5' hydroxyl group is protected by a dimethoxytrityl (DMT) group, while the 3' position carries a phosphoramidite function that is reactive toward coupling. Unlike the adenine, cytosine, and guanine monomers, thymine has no exocyclic amine, so this monomer requires no additional protecting group on its base. In the laboratory it serves as one of the four core monomers delivered by an automated DNA synthesiser, building an oligonucleotide strand step by step on a solid support through the repeating cycle of deprotection, coupling, capping, and oxidation.
The most notable characteristics of this material are the simplicity of its protection pattern and the reliability of its coupling reaction. Because the base itself does not need to be protected, thymidine monomers generally show clean and efficient synthesis behaviour, and they are frequently used as a benchmark monomer when researchers evaluate instrument performance or reagent quality. The dimethoxytrityl group also allows the efficiency of each individual cycle to be monitored through colorimetric measurement of the trityl cation released during detritylation. As with all phosphoramidites, quality is determined by low water content and freedom from contaminants that would interfere with the coupling step.

Handling and Storage
Store this phosphoramidite in a cool, dry location in its original tightly closed container, protected from moisture and atmospheric humidity, since water content is the principal factor that degrades coupling performance. Keep the reagent in a sealed, moisture-resistant vessel and allow the container to reach ambient temperature before opening to prevent condensation on the contents. Handle in a dry environment, ideally under an inert atmosphere, and use clean, dry glassware and anhydrous solvents when dissolving the material for use on a synthesiser. Work in a well-ventilated area or fume hood, wear gloves, safety glasses, and a laboratory coat, and consult the manufacturer's safety data sheet before handling.
