DPPH reagent (2,2-diphenyl-1-picrylhydrazyl) is a stable purple free radical compound used in spectrophotometric methods to test antioxidant activity; when it reacts with an antioxidant, its color changes to yellow and its absorbance decreases, indicating the sample's ability to neutralize free radicals. Working Principle of DPPH Reagent
The DPPH reagent works by trapping DPPH free radicals with antioxidant compounds through hydrogen atom or electron transfer. This converts DPPH into a yellow, non-radical reduced form, resulting in a decrease in the intensity of the purple color that can be measured using a spectrophotometer.

When a test sample containing antioxidants is added to the DPPH solution, hydrogen atoms from the antioxidant are donated to the DPPH radical. After the color change occurs, the DPPH reagent is measured using a UV-Vis spectrophotometer at a wavelength of 517 nm. A decrease in absorbance at this wavelength indicates high antioxidant activity.
