- Transferring microbial colonies from one medium to another.
- Streaking the agar surface to separate single colonies.
- Spreading the inoculum evenly across the agar surface (spreader).
Calibrated-volume loops are used when the amount of inoculum needs to be measured; wire loops are used when they will be flamed repeatedly. Disposable loops are discarded directly into biological waste containers; wire loops are flamed until red-hot before and after use.
