2-Fluoropyridine-3-carbonitrile (CAS 3939-13-7), also known as 2-fluoronicotinonitrile or 3-cyano-2-fluoropyridine, is a nicotinonitrile building block carrying a fluorine substituent at the 2-position of the pyridine ring. It is supplied as white crystals with a purity above 98%.
In pharmaceutical synthesis, this compound is a common starting material for active pharmaceutical ingredients (APIs), including niflumic acid, a drug used to relieve joint and muscular pain. It also provides a route to pyrazopyridine, a bicyclic heterocycle: hydrazine first displaces the fluorine by nucleophilic aromatic substitution, and the intermediate then undergoes pyrazole ring closure.
The pyridine core can be further functionalised by Minisci reaction to introduce alkyl groups at the meta and para positions. With several reactive functional groups on one small molecule, it is a versatile intermediate for drug discovery, medicinal chemistry and biochemistry research.
- Precursor for the synthesis of active pharmaceutical ingredients such as niflumic acid.
- Synthesis of pyrazopyridine bicyclic heterocycles via nucleophilic aromatic substitution with hydrazine and pyrazole cyclization.
- Meta- and para-alkylation of the pyridine ring through Minisci reaction.
- Building block for drug discovery, medicinal chemistry and biochemistry research.
