3-Fluoro-2-methylbenzoic acid (CAS 699-90-1), also called 3-fluoro-o-toluic acid, is a fluorinated benzoic acid building block supplied by Ossila as a white powder with purity above 98%. Structurally it derives from ortho-toluic acid (methylbenzoic acid), and its several functional groups make it a convenient intermediate for active pharmaceutical ingredients (APIs) and liquid crystal materials.
In medicinal chemistry, the compound serves as the starting material for acetoxy-methylbenzoic anhydride, a derivative showing antibiotic activity with a minimum inhibitory concentration (MIC) of 500–1000 μg/mL. It can also be converted into diarylmethanes, new building blocks for sodium-glucose transporter 2 (SGLT2) inhibitors, by Friedel-Crafts acylation and a subsequent reduction step.
Because the carboxylic acid and fluoride sit in a meta relationship on the ring, the molecule offers a useful scaffold for designing bent-core liquid crystals. It has a molecular weight of 154.14 g/mol and melts at 158 °C – 160 °C.
