4-Fluoro-2-methylphenol (CAS 452-72-2), also called 4-fluoro-o-cresol, is an o-cresol carrying a fluorine atom in the para position. Supplied by Ossila as pale yellow crystals with >98% purity, it is a versatile building block in medicinal chemistry, polymer modification and biochemistry research.
In drug discovery, this phenol is used as a starting material for active pharmaceutical ingredients. When incorporated into quinazolines in place of unsubstituted phenol, it provides selective inhibition of receptor tyrosine kinase and better hepatocyte stability.
The compound also serves in coordination chemistry: a thermal Mannich condensation with ethanolamine and formaldehyde gives a tetradentate aminophenol ligand. Its vanadium complex shows a ligand-to-metal charge transfer (LMCT) state lasting 423 ps, long enough to drive photochemical reactions. In addition, laccase-catalysed grafting onto natural fibres such as flax and coconut allows their hydrophobicity to be tuned.
- Building block for the synthesis of active pharmaceutical ingredients, including quinazoline-based kinase inhibitors.
- Preparation of tetradentate aminophenol ligands by Mannich condensation.
- Synthesis of long-lived LMCT vanadium complexes for photochemical reactions.
- Laccase-catalysed grafting onto flax and coconut fibres to modify hydrophobicity.
- Research on photoresist materials.
