4-Fluoroindole (4-Fluoro-1H-indole, CAS 387-43-9) is an indole carrying a single fluorine atom, in which a pyrrole ring is fused to a benzene ring. Supplied by Ossila as a white powder with purity above 98%, it serves as a building block for dyes and active pharmaceutical ingredients in drug discovery, OLEDs, dye-sensitized solar cells and bioimaging.
The indole core offers several handles for derivatization. The nitrogen at the 1-position can be substituted after treatment with sodium hydride or potassium hydride, which turns it into a nucleophile. The 3-position can be activated with a Grignard reagent such as methylmagnesium bromide or with an organozinc compound, while the 2-position is accessible through directed ortho lithiation. Extended-chromophore indole derivatives known as triazatruxenes have been used as donor units in dye-sensitized solar cells reaching 13.6% power conversion efficiency.
Because indole acts as an intercellular signalling molecule, it is an attractive scaffold for API design. 4F-Indole has been reported to boost the activity of the aminoglycoside antibiotic kanamycin against the multi-drug resistant pathogen Pseudomonas aeruginosa. Its combination of photophysical behaviour and bioactivity also supports bioimaging work, such as tracking cancer cells during anti-cancer treatment.
