5-Bromo-4-fluoro-2-methylaniline (CAS 627871-16-3) is a dihalogenated aniline building block for drug discovery, medicinal chemistry and biochemistry. Structurally it is 2-methylaniline carrying a fluorine atom at the 4-position and a bromine atom at the 5-position, supplied as a pale purple powder with >98% purity.
Its several functional groups give chemists more than one handle to work with. The bromide can take part in cross-coupling reactions, the amine is open to nucleophilic substitution, and the fluoride can undergo nucleophilic aromatic substitution, making the compound a convenient intermediate for active pharmaceutical ingredients (APIs).
A notable use is in preparing MDL compounds, N-phenyl-4-(phenylsulfonamido)benzenesulfonamide derivatives such as MDL-800, MDL-801 and MDL-811. These molecules activate sirtuin 6 (SIRT6), a lysine deacetylase with tumour-suppressor activity, by binding at an allosteric site; the enhanced deacetylase activity lowers histone levels in human hepatocellular carcinoma cells.
- Key starting material for synthesising the SIRT6 activators MDL-800, MDL-801 and MDL-811.
- Synthesis intermediate for active pharmaceutical ingredients.
- Substrate for cross-coupling reactions at the bromide position.
- Fluorinated building block for nucleophilic aromatic substitution in medicinal chemistry.
- Aniline building block for drug discovery and biochemistry research.
