Description

TCI D4229 Daptomycin is a cyclic lipopeptide antibiotic naturally produced by soil bacteria, recognised for its activity against Gram-positive bacteria, including strains that have become resistant to many other antibiotics. In the laboratory setting, this compound is supplied as a research reagent for microbiological testing, studies of antibiotic mechanisms of action, and antimicrobial resistance research. Its distinctive structure, consisting of a peptide ring with a lipid tail, gives it a mode of action different from that of beta-lactam and glycopeptide antibiotics, which makes it an important subject of investigation.

The characteristic property that makes daptomycin so widely studied is its dependence on calcium ions. The presence of calcium changes the conformation of the molecule so that its lipid tail can insert into the cell membrane of Gram-positive bacteria, causing membrane depolarisation and loss of membrane potential without first having to penetrate the cell wall. Because of this mechanism, daptomycin susceptibility testing must be carried out on media containing calcium at an appropriate concentration, and this requirement is precisely what makes the compound an attractive model for studying antibiotic–membrane interactions.

As a research reagent, daptomycin is typically used in Indonesian laboratories in microbiology and life science research settings, where work focuses on Gram-positive bacteria, antibiotic mechanisms, and antimicrobial resistance. It is handled in laboratory-scale quantities for susceptibility testing and mechanistic assays, with attention to the calcium content of the culture media used so that observed activity reflects the compound's true mode of action.

Laboratory Applications

  • Antimicrobial susceptibility testing — suitable because daptomycin activity against Gram-positive bacteria is well characterised, provided the test medium contains calcium at an appropriate concentration.
  • Antibiotic mechanism of action studies — its calcium-dependent insertion into the bacterial membrane makes it a clear model for investigating how antibiotics depolarise membranes.
  • Antimicrobial resistance research — it retains activity against strains resistant to many other antibiotics, making it useful for studying resistance patterns among Gram-positive isolates.
  • Antibiotic–membrane interaction studies — the lipid tail and peptide ring structure allow researchers to examine membrane targeting without prior penetration of the bacterial cell wall.
  • Comparative antibiotic class research — its mode of action differs from beta-lactam and glycopeptide antibiotics, so it serves as a reference compound when contrasting antibacterial mechanisms.

Specifications

  • Brand: TCI
  • CAS number: 103060-53-3
  • Category: Life Science > Biochemicals and Reagents > Antibiotics
  • Product code: D4229
  • Pack sizes: available in laboratory research pack sizes; please confirm the available packaging option when ordering
  • Storage: store according to the manufacturer's instructions stated on the product label and safety data sheet

Handling and Storage

Daptomycin should be stored under the conditions specified by the manufacturer on the product label and in the accompanying safety data sheet, in its original tightly closed container, protected from moisture and kept away from heat and direct sunlight. Keep the container closed when not in use and allow it to reach room temperature before opening to limit moisture uptake. Handle the material in a clean laboratory area using gloves, a laboratory coat, and eye protection, and avoid generating dust or aerosols when weighing. Prepare solutions with clean, appropriate labware, label all working solutions clearly, and consult the safety data sheet before use and before disposal of residues. As an antibiotic research reagent, it should be handled only by trained laboratory personnel.
