What is disodium beta-glycerophosphate pentahydrate and why does it matter?

Disodium beta-glycerophosphate pentahydrate (CAS 13408-09-8) is the sodium salt of a glycerol phosphate ester. The phosphate group is attached to the central carbon atom (the beta position) of glycerol. Its molecular formula is C₃H₇Na₂O₆P·5H₂O, with a molar mass of about 306.1 g/mol for the pentahydrate form. In the TCI catalog, it is available under product code D3964 as a grade for biochemical research. In practice, you will also find it under the names β-glycerophosphate, β-GP, or sodium β-glycerophosphate.

It matters because one simple molecule can play three different roles in the laboratory. The three roles are:

  • An organic phosphate source that enzymes release gradually.
  • A serine/threonine phosphatase inhibitor in lysis buffers and kinase assays.
  • A media component and weak buffer in certain cultures.

All three roles depend on purity and weighing accuracy, so the details of the raw material determine the results.

Properties that determine experimental outcomes

A phosphate ester, not free phosphate. β-GP is an ester. Inorganic phosphate is released only when an enzyme such as alkaline phosphatase (ALP) hydrolyzes it. This difference matters in osteogenic cultures. If the material contains a high level of free inorganic phosphate, calcium phosphate can precipitate in the medium without any involvement of cell activity.

Readily soluble in water. Concentrated stock solutions are easy to prepare with molecular biology grade or ultrapure water. For this reason, mistakes occur more often in the mass calculation than in dissolving the material.

Pentahydrate form. Five molecules of water of crystallization are included in the weighed mass. Use the molar mass of the pentahydrate (±306.1 g/mol), not that of the anhydrous form (±216.0 g/mol). For example, a 1 M stock solution requires about 306.1 g per liter. Using the wrong molar mass throws the final concentration off by about 30%, and this error often goes unnoticed when comparing data between labs.

Positional isomers. Glycerophosphate also exists as the alpha isomer. Material with a high fraction of the alpha isomer is not equivalent to pure β-GP, especially in enzymatic assays. Check the specification on the certificate of analysis.

Weak buffer. The phosphate monoester group provides mild buffering capacity around neutral pH. This property is used in media for lactic acid bacteria, which produce acid during growth.

Solution stability. Phosphate esters can hydrolyze slowly, especially if the solution is contaminated with microbes or phosphatase enzymes, or if it is heated repeatedly. This affects how you prepare and store stock solutions (see the handling section).

Laboratory uses and roles

1. Supplement for osteogenic differentiation media.

In cultures of mesenchymal stem cells or osteoblasts, β-GP is used together with ascorbic acid and dexamethasone. Its role is that of a phosphate donor. ALP on the cell surface cleaves β-GP into inorganic phosphate, which then forms mineral deposits with calcium that can be stained with alizarin red or von Kossa. Commonly used concentrations are in the low millimolar range (often around 10 mM, but follow your own protocol). Concentrations that are too high can trigger non-physiological mineralization.

2. Phosphatase inhibitor in lysis buffers and kinase assays.

In protein phosphorylation analysis (for example, phospho-protein western blots or in vitro kinase assays), β-GP inhibits serine/threonine phosphatases so that the phosphorylation status of the sample does not change during extraction. It is usually combined with other inhibitors, such as sodium orthovanadate or sodium fluoride, because its inhibition spectrum is limited. β-GP is not a replacement for a complete inhibitor cocktail.

3. Substrate or component of phosphatase assays.

Because it is hydrolyzed by ALP and other phosphatases, β-GP can serve as a substrate in phosphatase activity tests. The released phosphate is then measured with a colorimetric phosphate method.

4. Media and buffer component for microbes.

Certain media for lactic acid bacteria, such as M17-type formulations, use β-GP as a phosphate source and pH buffer. Its buffering role limits the drop in pH caused by lactic acid production, so growth continues for longer.

5. Applications outside biology.

Recent research shows that this compound is also being studied in mineral flotation (see the next section).

What does recent research say?

Here are two relevant studies.

  • Growth of *Streptococcus thermophilus*. The study *Investigating the Role of β-Disodium Glycerophosphate and Urea in Promoting Growth of Streptococcus thermophilus from Omics-Integrated Genome-Scale Models* (Foods, 2024) examines the role of β-GP and urea in supporting the growth of this bacterium, using an omics-integrated genome-scale model approach. For food microbiologists, this article shows that β-GP in media is not just a filler, but a component whose metabolic and buffering roles deserve consideration. DOI: 10.3390/foods13071006.
  • Flotation of copper oxide ore. The study *Separation mechanism of malachite and dolomite using a novel depressant disodium glycerophosphate in copper oxide ore flotation* (Surfaces and Interfaces, 2026) investigates disodium glycerophosphate as a novel depressant in separating malachite from dolomite. This shows that glycerophosphate salts also interest mineral processing researchers, not only biochemists. For research of this kind, purity and batch consistency still matter. Why does TCI quality matter for this material?

TCI is the manufacturer, and AMI Scientific is the official TCI distributor in Indonesia. Here is what to look for and how it affects your experiments.

Purity and impurity content. Free inorganic phosphate is the most harmful impurity in osteogenic cultures, because it triggers calcium phosphate precipitation and gives false mineralization signals in staining. In phosphatase assays, free phosphate raises the background (blank) signal and lowers sensitivity. Metal impurities can affect metal-sensitive enzymes. For this reason, choose a grade intended for biochemical research such as D3964, especially for cell culture and enzymatic assays.

Batch-to-batch consistency. Osteogenic differentiation experiments run for weeks. If the material batch changes in the middle of a series of experiments and the impurity or water content differs, the rate of mineralization can shift and data between replicates become hard to compare. Recording the lot number in your lab notebook is a simple step that helps greatly when tracing the cause of variation.

Analytical documentation. The certificate of analysis (CoA) and SDS let you check purity, identity, and relevant parameters such as isomers or water content before use. For QC analysts and labs that work with audits, these documents are part of traceability. For non-biochemical work, such as rough buffer preparation or preliminary tests, another grade such as TCI G0097 can be considered, as long as its specifications meet your needs. Compare the specifications of the two before deciding.

Packaging. The pentahydrate can absorb or release moisture if the container is opened often. Changes in water content shift the effective mass per gram. Tightly sealed packaging in a size that matches your usage helps keep the water content stable and reduces the risk of contamination from spatulas or lab air. For infrequent use, a small size makes more sense than one large container that is opened repeatedly.

Handling and storage

  • Store in the original tightly closed container, in a cool, dry place, protected from light and moisture. Follow the temperature stated on the label and in the TCI SDS.
  • Weigh quickly and close the container again immediately. Use a clean, dry spatula. Do not return leftover material to the original container.
  • Always calculate with the molar mass of the pentahydrate (±306.1 g/mol). Example: a 100 mM stock solution means about 30.6 g per liter.
  • Stock solutions for cell culture should be sterilized with a 0.22 µm filter, not autoclaved, because phosphate esters can be hydrolyzed by heat. Divide into small aliquots and store frozen to avoid repeated freeze-thaw cycles. Use your lab's protocol as the final reference.
  • Safety: wear gloves, a lab coat, and safety glasses. Read the SDS before use and dispose of waste according to your institution's laboratory procedures.

Quick FAQ

Is β-glycerophosphate the same as inorganic phosphate?

No. β-GP is an organic phosphate ester. Its phosphate becomes available only after an enzyme such as ALP cleaves it. That is why it is used in osteogenic cultures.

Is β-GP enough as the only phosphatase inhibitor?

Usually not. β-GP inhibits serine/threonine phosphatases, but its spectrum is limited. Combine it with other inhibitors according to your target.

Why must I use the pentahydrate molar mass?

Because the weighed material contains five molecules of water of crystallization. Using the anhydrous molar mass makes the final concentration lower than planned.

What is the difference between D3964 and G0097?

Both are material with CAS 13408-09-8. D3964 is intended for biochemical research, while G0097 is another grade. Check the specification and CoA of each before choosing according to your application.

Conclusion

Disodium beta-glycerophosphate pentahydrate is chemically simple, but experimental results depend heavily on its purity, water content, and the accuracy of your calculations. For biochemical research and cell culture, TCI D3964 is the option intended for those purposes. You can also compare it with TCI G0097 if your needs are different. Contact AMI Scientific for CoA and SDS documents and to discuss specifications before you start a series of experiments.
