N,N,N',N'-Tetramethyl-1,4-phenylenediamine Dihydrochloride from TCI is the dihydrochloride salt of TMPD. TMPD is a derivative of p-phenylenediamine in which each of the two nitrogen atoms carries two methyl groups. The compound is well known as an electron donor that oxidises easily into a deep blue radical cation known as Wurster's Blue. In the laboratory, this behaviour makes it a very useful oxidase test reagent in microbiology, a redox mediator in biochemistry, and a model compound in electrochemistry.

Chemists and microbiologists choose this material because its one-electron oxidation is fast and reversible, and it produces a colour change that is easy to see by eye or measure by spectrophotometry. In the bacterial oxidase test, TMPD passes electrons to cytochrome c and turns blue-purple when the organism has cytochrome c oxidase. The dihydrochloride salt is more stable and dissolves more readily in water than the free base, so fresh reagent solutions are easy to prepare. Even so, the material remains sensitive to air and light.

In Indonesia, this reagent is widely used in clinical microbiology, food microbiology, animal health, and fisheries laboratories, where the oxidase test is a routine step in identifying bacterial isolates. University teaching and research laboratories use it in biochemistry courses and in studies of electron transport. Electrochemistry groups use it as a well-characterised reversible redox couple when they test electrodes and measurement setups.

  • Bacterial oxidase test: TMPD donates electrons to cytochrome c, giving a clear blue-purple colour that separates oxidase-positive organisms from oxidase-negative ones during routine identification.
  • Redox mediation in biochemistry: its fast, reversible one-electron oxidation lets it shuttle electrons between enzymes and acceptors, which helps when studying respiratory chain components and cytochrome activity.
  • Electrochemical model studies: the reversible formation of the Wurster's Blue radical cation makes it a convenient reference system for checking electrodes, cells, and voltammetric procedures in research laboratories.
  • Spectrophotometric assays: the intensely coloured oxidised form can be followed with a spectrophotometer, so analysts can measure oxidation reactions instead of relying only on visual judgement.
  • Teaching demonstrations: the quick and visible colour change clearly shows electron transfer and radical cation formation, which makes it well suited to practical classes in microbiology, biochemistry, and physical chemistry.

Storage sensitive to air and light; keep tightly sealed and protected from light
  • Form: dihydrochloride salt of TMPD, more water-soluble than the free base

Because this material is sensitive to air and light, keep it in its original tightly closed container, ideally amber or opaque, in a cool, dry place away from direct light and oxidising agents. Reseal the container promptly after each use to limit exposure to air and moisture. Oxidase reagent solutions should be prepared fresh, because the dissolved compound gradually oxidises and darkens on standing. Follow the TCI safety data sheet, wear gloves, eye protection, and a lab coat, avoid creating dust, and handle the material in a well-ventilated area or fume hood. Dispose of waste according to local laboratory and environmental regulations.
