Description

5-Ethynyluridine, commonly abbreviated as EU, is a uridine nucleoside analogue that carries an ethynyl group at the 5 position of the uracil ring. The compound is a key reagent in molecular biology research for labelling newly synthesised RNA inside living cells. Its working principle is simple yet elegant: cells supplied with EU incorporate the analogue into RNA that is actively being transcribed, and the ethynyl group embedded within that RNA is then detected through a click reaction with an azide-labelled dye. The result is a picture of transcriptional activity that can be visualised directly.

The advantages that make EU the reagent of choice lie in the very small size of the ethynyl group, which keeps interference with biological processes to a minimum, and in the selectivity of the click reaction, which barely touches other functional groups present in the cell. This approach replaces older methods based on radiolabelled nucleosides or antibodies, because it requires no handling of radioactive material and demands no harsh denaturation to open access for the probe. Click-based detection also produces sharp signals with low background, making it suitable for both fluorescence microscopy and flow cytometry analysis.

In Indonesian laboratories, EU is typically used in university research groups, biomedical and molecular biology institutes, and cell culture facilities that study gene expression and transcription dynamics. It fits workflows where fluorescence microscopy or flow cytometry is already established, and where investigators need a non-radioactive route to visualising nascent RNA. Supplied by TCI under catalogue number E1560, it is handled alongside the usual click chemistry reagents such as azide-labelled fluorescent dyes.

Laboratory Applications

  • Nascent RNA labelling in cultured cells: EU is incorporated into RNA during active transcription, allowing newly synthesised transcripts to be distinguished from the pre-existing RNA pool without radioactive tracers.
  • Transcriptional activity imaging by fluorescence microscopy: the click reaction with azide-labelled dyes yields sharp signals and low background, so sites of active transcription can be visualised directly within the cell.
  • Flow cytometry analysis of transcription: click-based detection of EU-labelled RNA gives clean, quantifiable fluorescence across large cell populations, supporting comparison of transcriptional activity between samples.
  • Click chemistry and conjugation chemistry method development: the ethynyl handle reacts selectively with azide partners and barely touches other cellular functional groups, making EU a dependable model substrate for protocol optimisation.
  • Gene expression studies as an alternative to antibody or radiolabel methods: no radioactive handling is required and no harsh denaturation is needed to give the probe access to the target RNA.

Handling and Storage

Store the material in its original tightly closed container, following the storage conditions stated on the manufacturer's label and safety data sheet. Keep the container sealed and protected from moisture, and return it promptly to the designated storage location after each use. Weigh and dispense in a clean, well-ventilated working area, and prepare solutions using appropriate solvent and labware for cell culture work. Wear standard laboratory personal protective equipment, including gloves, a laboratory coat, and eye protection, and avoid direct contact with skin, eyes, and clothing. Use only clean, dry spatulas to prevent contamination of the remaining stock, and record the opening date on the container. Dispose of residues and contaminated consumables in accordance with the applicable laboratory chemical waste procedures. Consult the manufacturer's safety data sheet before first use.
