Description

Di(N-succinimidyl) Sebacate is a homobifunctional crosslinking reagent carrying two N-hydroxysuccinimide (NHS) ester groups at either end of a ten-carbon sebacate chain. The NHS ester groups react with primary amines — principally the amino groups of lysine residues and the N-terminus of a protein chain — to form stable amide bonds. Because both ends of the molecule are reactive toward the same functional group, the reagent is used to join two protein-bearing molecules together, to immobilise biomolecules onto solid surfaces, or to lock protein–protein interactions in place so that they can be analysed further in the chemical biology laboratory.

The advantage that makes this reagent a preferred choice lies in the length of its spacer arm. The sebacate chain provides a more generous distance between the two points of attachment than short-arm crosslinkers do, which makes it well suited to bridging amine groups that are located far apart from one another, or to keeping an immobilised biomolecule free enough to move so that it does not lose its function. That long aliphatic chain is hydrophobic in character and dissolves well in organic solvents such as dimethyl sulfoxide or dimethylformamide, so stock solutions are normally prepared in one of those solvents before being added to the reaction.

In Indonesian laboratories, this type of reagent is typically handled in university research groups, biotechnology institutes, and protein chemistry facilities working on conjugation and bioimmobilisation. The usual working pattern is to prepare a concentrated stock in dimethyl sulfoxide or dimethylformamide first, then introduce a small volume of that stock into the aqueous protein solution being crosslinked. Because the reagent is supplied by TCI and is used in small quantities per experiment, it is normally kept as a shared stock item within the laboratory.

Handling and Storage

Store the container tightly sealed in a cool, dry place and protect it from moisture, because the NHS ester groups are hydrolysed by water and lose reactivity once that happens. Keep the reagent in its original tightly closed container, and allow it to reach ambient temperature before opening so that condensation does not form inside. Prepare stock solutions in dry dimethyl sulfoxide or dimethylformamide immediately before use rather than storing dilute aqueous solutions.
