TCI N0619 4-Nitrophenyl alpha-D-Mannopyranoside is a chromogenic substrate for the enzyme alpha-mannosidase. It consists of a mannose sugar attached to 4-nitrophenol through an alpha-glycosidic bond. When alpha-mannosidase cleaves this bond, 4-nitrophenol is released. Under alkaline conditions it turns yellow, and the colour can be measured by spectrophotometry. In glycobiology and biochemistry laboratories, this substrate is used to measure alpha-mannosidase activity from many sources, including animal tissues, plants and microorganisms.
Researchers choose this substrate because it is specific to the alpha-mannosidic bond and gives results that are easy to quantify. Alpha-mannosidase plays a key role in glycoprotein processing, where it trims sugar chains that determine how proteins function and where they go inside the cell. A deficiency of the lysosomal form of this enzyme is also linked to lysosomal storage disorders. Because the readout is a simple colour change, researchers can study the enzyme without complex equipment. A spectrophotometer or microplate reader, which most laboratories already have, is enough to follow the reaction and compare activity between samples.
In Indonesia, this substrate is useful for biochemistry laboratories at universities, research institutes and life science facilities that study enzymes and carbohydrate metabolism. Teams working on glycoscience, enzyme characterisation from local plants or microbes, and basic biomedical research can use it to set up simple and reproducible alpha-mannosidase assays. Its straightforward colorimetric readout also makes it suitable for teaching laboratories, where students learn the principles of enzyme kinetics and substrate specificity with standard instruments.

- Alpha-mannosidase activity assays: the substrate releases 4-nitrophenol when cleaved, producing a yellow colour in alkaline conditions that can be measured directly by spectrophotometry.
- Enzyme characterisation from biological sources: researchers can compare alpha-mannosidase activity in animal tissues, plants and microorganisms using one consistent, specific chromogenic substrate.
- Glycoprotein processing research: because alpha-mannosidase trims sugar chains on glycoproteins, this substrate helps scientists study the enzymes that control protein function and localisation.
- Lysosomal storage disorder studies: deficiency of lysosomal alpha-mannosidase is linked to these disorders, so a simple activity assay supports basic research into enzyme function.
- Microplate-based screening: the colorimetric readout suits microplate readers, so many samples or conditions can be tested in parallel with standard laboratory equipment.

Store 4-Nitrophenyl alpha-D-Mannopyranoside in its original, tightly closed container, following the storage temperature on the TCI label. Keep it in a dry place away from moisture, heat and direct light to protect the substrate before use. Once opened, reseal the container promptly and avoid repeated exposure to humid air. Handle the material in a clean, well-ventilated area while wearing standard personal protective equipment, including a lab coat, gloves and safety glasses. Avoid creating dust or letting it touch your skin or eyes. Prepare substrate solutions fresh when possible, and review the Safety Data Sheet before handling and disposal.
